The development of antiresorptive agents for osteoporosis has been hampered by the lack of an appropriate model system for bone biology. We have developed a quantitative assay for measuring transcriptional activation of the human osteocalcin (OC) gene. This assay was used to screen candidate factors for OC promoter activation. We found that bone-derived human tissue-nonspecific alkaline phosphatase (TNAP) induced transcriptional activation of the OC gene in a cell-free assay using HeLa nuclear extracts. Since TNAP is a major membrane-bound bone-derived ectoenzyme and produces a peptide that shares sequence homology with the amino-terminus of human OC, we synthesized a peptide corresponding to the amino-terminus of OC (POC) and tested its ability to activate the OC promoter in the HeLa nuclear extract-based OC gene transcriptional activation assay. Although the synthetic POC peptide alone did not activate the OC promoter, it activated the OC promoter when co-incubated with TNAP. The effect was concentration-dependent and was blocked by a specific POC-derived OC-cDNA.
